Hormonal regulation of regucalcin gene expression: involvement in cell metabolism
نویسنده
چکیده
Regucalcin (RGN/SMP30) and its gene (rgn)are identified in over 15 species consisting of regucalcin family, and its gene is localized on X chromosome. Regucalcin has been shown to play a multifunctional role in many tissues and cell types. Hormonal regulations of regucalcin gene expression are mediated through various transcription factors (including AP-1, NF1-A1, RGPR-p117 and other), which are involved in phosphorylation and dephosphorylation by intracellular signaling factors. Regucalcin mRNA expression has been shown to be regulated by hormonal factors (including calcium, calcitonin, parathyroid hormone, 1,25-dihydroxyvitamin D3, insulin, tumor necrosis factor-α, transforming growth factor-β, estrogen, testosterone, aldosterone, dexamethasone and other) in various tissues and many cell types. Regucalcin mRNA expression has also been shown to suppress with pathophysiological states that are related to hormonal diseases. Regucalcin may be a key molecule that is involved in hormonal effect and its related metabolic disease.

Regucalcin, which was discovered as a calcium-binding protein in 1978, has been demonstrated to play a multifunctional role as a suppressor in signaling transduction in various types of cells and tissues. Regucalcin has been shown to regulate intracellular calcium homeostasis, protein kinases and phosphatases, nitro oxide production, protein synthesis, gene expression, cell proliferation and ap...
متن کاملRegucalcin, a calcium-binding protein, is a new target gene in human prostate cancer?
Regucalcin was identified as a calcium (Ca)-binding protein playing an important role in maintenance of intracellular Ca homeostasis. More recently, proteomic studies have identified it as a down-regulated gene in mouse and human hepatocellular carcinomas. In addition, regucalcin effects in kidney and liver cells, suppressing cell proliferation, inhibiting expression of oncogenes, and increasin...
متن کاملThe potential role of regucalcin in kidney cell regulation: Involvement in renal failure (Review).
The kidneys play a physiologic role in the regulation of urine formation and nutrient reabsorption in the proximal tubule epithelial cells. Kidney development has been shown to be regulated through calcium (Ca2+) signaling processes that are present through numerous steps of tubulogenesis and nephron induction during embryonic development of the kidneys. Ca2+-binding proteins, such as calbindin...
متن کاملHormonal regulation of regucalcin mRNA expression in osteoblastic MC3T3-E1 cells.
The effect of various hormones on regucalcin mRNA expression in osteoblastic MC3T3-E1 cells in vitro was investigated. Cells with subconfluency were cultured for 24 or 48 h in a medium containing either vehicle or various hormones without fetal bovine serum. Regucalcin mRNA expression was significantly increased after culture with parathyroid hormone (synthetic human PTH; 10(-7) M), insulin-lik...
